Crop Genetic Resources: An Economic Appraisal AgEcon
Day-Rubenstein, Kelly A.; Heisey, Paul W.; Shoemaker, Robbin A.; Sullivan, John; Frisvold, George B..
Crop genetic resources are the basis of agricultural production, and significant economic benefits have resulted from their conservation and use. However, crop genetic resources are largely public goods, so private incentives for genetic resource conservation may fall short of achieving public objectives. Within the U.S. germplasm system, certain crop collections lack sufficient diversity to reduce vulnerability to pests and diseases. Many such genetic resources lie outside the United States. This report examines the role of genetic resources, genetic diversity, and efforts to value genetic resources. The report also evaluates economic and institutional factors influencing the flow of genetic resources, including international agreements, and their...

Cryopreservation of coffee zygotic embryos: dehydration and osmotic rehydration Ciência e Agrotecnologia
Pinto,Maísa de Siqueira; Paiva,Renato; Silva,Diogo Pedrosa Corrêa da; Santos,Paulo Augusto Almeida; Freitas,Rodrigo Therezan de; Silva,Luciano Coutinho.
ABSTRACT Conservation of plant genetic resources is important to prevent genetic erosion. Seed banks are the most common method of ex situ conservation; however, coffee seeds can not be stored by conventional methods. Cryopreservation is a viable alternative for long-term conservation of species that produce intermediate or recalcitrant seeds, as coffee. The aim of this work was to cryopreserve Coffea arabica L. cv Catuaí Vermelho IAC 144 zygotic embryos, and analyse the effects of dehydration prior cryopreservation and osmotic rehydration after thawing, in embryos germination and seedlings formation after cryopreservation. Prior to cryopreservation, different dehydration times (0, 15, 30, 60 and 120 min) were tested. Dehydrated embryos were cryopreserved...

In vitro conservation of blackberry genotypes under minimal growth conditions and subsequent large-scale micropropagation PAB
Gomes,Hugo Teixeira; Bartos,Patrícia Monah Cunha; Andrade,Maíra Teixeira de; Almeida,Raphael Ferreira; Lacerda,Luciana Florencio de; Scherwinski-Pereira,Jonny Everson.
Abstract: The objective of this work was to evaluate the micropropagation of blackberry (Rubus spp.) cultivars, after in vitro conservation under minimal growth conditions. Nodal segments of the 'Guarani', 'Caingangue', 'Ébano', and 'Xavante' genotypes were conserved under minimal growth conditions at 20ºC, for 15 months. Microshoots were regenerated and multiplied by up to five successive subcultures, when they were rooted and acclimatized. After 30 days of acclimatization in a greenhouse, rooted plantlets showed no significant losses. Blackberry cultivars can be conserved in vitro for 15 months, without subcultures and, after this time, they can be micropropagated on a large-scale, maintaining the regenerative potential and multiplication.

In vitro conservation of Piper aduncum and Piper hispidinervum under slow-growth conditions PAB
Silva,Tatiane Loureiro da; Scherwinski-Pereira,Jonny Everson.
The objective of this work was to evaluate in vitro storage of Piper aduncum and P. hispidinervum under slow-growth conditions. Shoots were stored at low temperatures (10, 20 and 25°C), and the culture medium was supplemented with osmotic agents (sucrose and mannitol - at 1, 2 and 3%) and abiscisic acid - ABA (0, 0.5, 1.0, 2.0 and 3.0 mg L-1). After six-months of storage, shoots were evaluated for survival and regrowth. Low temperature at 20ºC was effective for the in vitro conservation of P. aduncum and P. hispidinervum shoots. In vitro cultures maintained at 20ºC on MS medium showed 100% survival with slow-growth shoots. The presence of mannitol or ABA, in the culture medium, negatively affected shoot growth, which is evidenced by the low rate of...

Is the genetic integrity of natural plant populations ex situ preserved with the current sampling, conservation and regeneration approaches? JBAG
Camadro,E.L.
Acceleration of environment degradation and climate change poses serious threats to both natural and agricultural ecosystems. In fact, the number of endangered plant species at the global level has increased yearly in the last decades, due to anthropic interventions that sum up to the climate change. Samples of natural plant populations, particularly of wild crop relatives, are ex situ conserved in germplasm banks, which have become active in the provision of genetic resources to breeders. The aim of this article is to generate discussion on the adequacy of (a) current protocols for sampling and ex situ conservation of the natural genetic diversity and (b) the actual species paradigm in plants, due to their consequences for this and future generations.

Orchid cryopreservation Ciência e Agrotecnologia
Vendrame,Wagner; Faria,Ricardo Tadeu de; Sorace,Mauren; Sahyun,Sandra Aparecida.
Orchids are lush and highly valuable plants due to their diversity and the beauty of their flowers, which increases their commercialization. The family Orchidaceae comprises approximately 35,000 species, distributed among more than 1,000 distinct genera and 100,000 hybrids, totaling approximately 8% to 10% of all flowering plants. With the advance of agriculture and the constant destruction of their natural habitat, orchid species are collected in an indiscriminate manner by collectors and vendors, and this extractive activity threatens many species with extinction, drastically reducing their genetic variability in nature. Therefore, it is essential to seek alternatives that make the preservation of such species feasible using techniques with low...

Tropical seed species' responses to liquid nitrogen exposure Braz. J. Plant Physiol.
Salomão,Antonieta Nassif.
The ability to tolerate ultra-low temperature (-196 ºC) exposure was evaluated in 66 tropical orthodox seed species of 21 botanical families from the Cerrado (Brazilian Savannah) and Atlantic Forest Brazilian biomes. Liquid nitrogen had no effect on the germinability of 51 seed species. The stimulatory effect of cryogenic temperature on germinability, with or without subsequent chemical scarification, was observed in nine seed species with deep physical dormancy, or heterogeneous levels of seed hardness, or with no dormancy. Significant reduction in germinability occurred in six seed species, presumably because of factors acting individually or in combination on these seeds, such as inappropriate moisture content, or the potentially detrimental effect of...

DEMARCHI,GRASIELA; STEFENON,VALDIR M.; STEINER,NEUSA; VIEIRA,FELIPE N.; VESCO,LIRIO L. DAL; GUERRA,MIGUEL P..
This study aimed to establish a cryopreservation protocol for embryogenic cultures of A. angustifolia, enabling the ex situ conservation of the species. Embryogenic cultures were established from immature seeds and treated with variations of the cryoprotectant solutions SuDG, SoD and PVS2 prior to immersion in liquid nitrogen. Cell viability was evaluated after 30, 60 and 90 days of re-growth. The highest re-growth without morphological alterations and with normal biochemical composition was obtained with the PVS2 solution with 40 min immersion in ethanol (-20 °C). This procedure opens new horizons for the ex situ conservation of the species genetic.
